چکیده

1. Pea-seed phosphofructokinase was purified 27-fold by a combination of fractionation with ethanol and ammonium sulphate. Under the conditions of assay, the enzyme was strongly inhibited by phosphoenolpyruvate. This inhibition was reversed by increasing the concentration of fructose 6-phosphate or magnesium chloride, or by lowering the ATP concentration. 2. Citrate, ADP and AMP inhibited phosphofructokinase and increased the sensitivity to phosphoenolpyruvate inhibition. Sulphate and inorganic phosphate stimulated the enzyme activity and decreased the sensitivity to phosphoenolpyruvate. 3. In the presence of inorganic phosphate and low concentrations of ATP, inhibition by phosphoenolpyruvate ceased and phosphoenolpyruvate became stimulatory. 4. The possible significance of these results in the control of plant carbohydrate metabolism is discussed.

Fructose 2,6-bisphosphate and the regulation of pyrophosphate-dependent phosphofructokinase activity in germinating pea seeds.

THE ACTIVITY OF PYROPHOSPHATE: d-fructose-6-phosphate-1-phosphotransferase (EC 2.7.1.90, PPi-PFK) in cotyledons and sprouts of germinating pea seeds (Pisum sativum cv Alaska or Green Arrow) increases rapidly during the first 2 to 3 days after imbibition and then declines to a lower activity. The reaction toward fructose 1,6-bisphosphate formation is activated greatly by fructose 2,6-bisphosphat...

Dry Matter Intake and Digestibility in Weast African Dwarf Goats Fed Raw or Processed Pigeon Pea (Cajanus Cajan) Seed

Four West African Dwarf (WAD) grower bucks averaging 6 kg in weight and aged between 5-6 months of age were used in a digestive trial to determine the intake and digestibility of raw or processed pigeon pea seed meal based diets. The diets designated RPBD, SPBD, TPBD and BPBD were formulated from cassava peel, palm kernel cake, brewers dry grain, bone meal and common salt to contain 15% of raw,...

Inactivation of pea leaf phosphofructokinase by light and dithiothreitol.

When intact pea (Pisum sativum L.) plants are illuminated, the glycolytic enzyme phosphofructokinase is inactivated. In crude extracts the enzyme is inhibited by dithiothreitol. It would seem that this cytoplasmic enzyme, like glucose-6-P dehydrogenase, is light-inactivated when the enzymes of photosynthetic carbon metabolism are light-activated.

Chloroplast Phosphofructokinase: II. Partial Purification, Kinetic and Regulatory Properties.

Chloroplast phosphofructokinase from spinach (Spinacia oleracea L.) was purified approximately 40-fold by a combination of fractionations with ammonium sulfate and acetone followed by chromatography on DEAE-Sephadex A-50. Positive cooperative kinetics was observed for the interaction between the enzyme and the substrate fructose 6-phosphate. The optimum pH shifted from 7.7 toward 7.0 as the fru...

Nutrient Intake and Digestibility by West African Dwarf (WAD) Sheep Fed Graded Levels of Pigeon Pea Seed Meal

A study was carried out to evaluate nutrient intake and digestibility by West African Dwarf (WAD) sheep fed graded levels of dietary pigeon pea seed meal (PSM). Four diets designated A, B, C and D were formulated to contain 0, 10, 20, and 30% levels of PSM, respectively. Four WAD rams aged between 15 and 17 months and weighing between 16 and 19 kg were used to conduct digestibility study in a 4...
